Comparison of the specificity of olive reactive IgE antibodies from patient groups with different geographical origin

ABSTRACTOlive tree (Olea europaea) pollen is an important cause of IgE-mediated allergy in the countries of the Mediterranean area. Despite the fact that olive trees are not grown in many other parts of the world such as north Europe and North America, there are people also in these areas who have IgE antibodies reactive to olive pollen extract.The aim of this study was to compare the specificity of olive pollen reactive IgE antibodies in serum samples from Spanish patients, who had been exposed to olive pollen and had an olive allergy diagnosis, and serum samples from patients with high IgE levels from Sweden and North America without any known exposure to olive pollen.Methods: The serum samples were analysed by ImmunoCAPTM (Phadia, Uppsala) for IgE antibody reactivity to olive pollen extract and Ole e1 (the main allergenic component in olive pollen).The reactivity to important allergens and allergenic components of trees (birch), grass (timothy) and weeds (wall pellitory) and cross reactive carbohydrate determinants (CCD) was also tested. Selected serum samples were analysed by Western blot using olive pollen extracts as antigen source and also by inhibition analysis of the binding of IgE antibodies to pollen extract in ImmunoCAPTM.Results: Most serum samples from the Spanish patients showed a strong response to olive pollen extract and to Ole e1 in both ImmunoCAPTM and immunoblotting analysis. The reactivity with birch, timothy and wall pellitory was in general weak or absent when evaluated by ImmunoCAPTM analysis and IgE antibody binding inhibition assay.
